I have two HDHomeRun EXTEND on my network and I would like to "reserve" one for Jellyfin use and one for HDHomeRun app. Is there a way I can do that? Currently when I start HDHomeRun app on my firestick, it detects both EXTEND and my understand is that it may use any of the tuners. On Jellyfin configuration, I am able to point it to one of the EXTEND and it will only use that.

There is no reservation. The official apps detect all devices and engines on the same subnet and the device can be used by any app on the same network at any time and most apps today will use "auto" to take any available tuner from a particular device. If you want a particular device to be used exclusively you could often directly attach a device to a "computer" that runs only that app and not other apps that may wish access for such as the device wouldn't be seen by anything else as it isnt on the network.
Another possibility is that the app should use a record engine if such is detected. You might be able to put the record engine on a device that is prevented from communicating with a particular tuner (i.e. through firewall rule). Then whenever the app requests a channel it should use the record engine that doesn't know about that other device.
Personally I wouldn't bother and only use apps that play nicely with sharing devices.
